Phenibut is the structural analogue of GABA with a phenyl ring addition at the C3 position that can penetrate the brain, unlike GABA itself. The phenyl functional group is lipophilic (fat-loving or water-abhorring, whichever you prefer). The lipophilicity of the phenyl moiety allows the molecule to pass freely into the phospholipid bilayer and enter the brain.
This same pharmacological strategy was employed in the development of other GABA analogues, like gabapentin (Neurontin), a drug approved in US for the treatment of neuropathic pain and epilepsy.
